ChemTest^TM Staph – Coagulase Test as lyophilized rabbit plasma EDTA is designed for the rapid identification of coagulase positive staphylococci

Method

Coagulase test is used for the identification of pathogenic Staphylococci, principally S. aureus by means of the coagulase test. When the strain is placed in contact with rabbit plasma, the appearance of coagulation is observed.

Preparation Coagulase Test

# Vial 40 tests: Add 12 ml of sterile distilled water, Mix well

# Vial 25 tests: Add 7.5 ml of sterile distilled water, Mix well

# Vial 10 tests: Add 0.3 ml of sterile distilled water, Mix well

Sample: Staphylococci colonies isolated on agar medium

Technique:

• Step 1: Transfer some Staphylococci colonies into Brain Heart Infusion Broth-BHIB; incubate 18-24h at 37⁰C.
• Step 2: Mix 0.3ml of Coagulase Test with 0.1ml of BHIB and incubate at 37⁰C and read the result after 3h, 6h. The test is negative after 24h.
• Positive: appearance of a clot within 24h
• Negative: Absence of a clot within 24h

Quality Control:

# Staph aureus ATCC 25923: Positive

# Staph epidermidis ATCC 12228: Negative

Storage:

• Stored at 2-8⁰C in the box and use before expiry date.
• Coagulase Test prepared: Stored at 2-8⁰C and use within 1 week.
