A selective medium useful for the isolation and identification of E.coli, Coliform with membrane filtration method of water samples Acc ISO 9308-1:2014

Preparation

# Dissolve 27g in 1000ml of distilled water (purified water)

# Heat and frequent stirring to completely dissolve the medium

# Heat with frequent stirring until boiling (DO NOT AUTOCLAVE)

# Cool to 45-50°C, mix well and dispense into appropriate sterile containers

Description

ChromoGel™ Coliform Agar is a selective medium for the isolation and identification of E.coli, Coliform with membrane filtration method of water samples acc ISO 9308-1:2014

• The medium can use as secondary selective agar plate on ISO method, petri-film method.
• The samples can streak directly on surface agar plates without enrichment step.

Technique

Membrane filtration method: ISO 9308-1

1. Filter the samples on a 0,45μ membrane and settle the last over the medium surface.
2. Tightly close the plate and incubate at 36 ± 2°C for 18-24 hours
3. Count E. coli and Coliform

Quality Control acc. to ISO 11133

• Productivity control at 37 ± 2ºC / 18-24H

• Selective control at 37 ± 2ºC / 18-24H

Storage

Dehydrate culture media powder keep at 2ºC to 30 ºC

Ready to use plates keep at 2ºC to 20 ºC

Use before the expiry date on the label

ChromoGel™ Coliform Agar là môi trường chọn lọc dùng trong nuôi cấy định danh e.coli và coliforms bằng phương pháp màng lọc theo ISO 9308-1

Chuẩn bị

# Hòa tan 27g trong 1000ml nước cất (hoặc nước đã lọc)

# Gia nhiệt và khuấy thường xuyên để môi trường hòa tan hoàn toàn

# Khử trùng ở 121° C trong 15 phút

# Lắc đều và rót vào các dụng cụ chứa vô trùng thích hợp

ChromoGel™ Coliform Agar là môi trường chọn lọc dùng trong nuôi cấy định danh e.coli và coliforms bằng phương pháp màng lọc theo ISO 9308-1

• Môi trường được dùng như đĩa thạch đối chứng thứ 2 dựa theo phương pháp ISO hoặc Petri-film
• Mẫu kiểm có thể được cấy trực tiếp lên bề mặt đĩa thạch không cần qua tăng sinh.

Phương pháp kỹ thuật

Phương pháp màng lọc theo ISO 9308-1

1. Lọc mẫu bằng mang lọc có kích thước lỗ lọc 0.45μm và đặt màng lọc đã lọc lên mặt thạch.
2. Ủ hiếu khí ở 37 ± 2ºC / 18-24H
3. Đếm khuẩn E.coli và Coliform

Đánh giá chất lượng theo ISO 11133

• Đánh giá hiệu năng ở 37 ± 2ºC / 18-24H

• Đánh giá chọn lọc 37 ± 2ºC / 18-24H

Bảo quản

Môi trường bột được bảo quản ở 2ºC đến 30 ºC

Đĩa thạch chuẩn bị sẵn ở 2ºC đến 20 ºC

Sử dụng trước hạn dùng ghi trên nhãn
